ReNTox
Backstory and manufacturer
ReNTox is a product of Pharma Research Bio Co., Ltd. (formerly BioCND) from South Korea, part of the Pharma Research Products (PRP) group [1][2]. In 2018, PRP acquired a majority stake in the ReNTox manufacturing plant (located in Gangneung, South Korea) and renamed it Pharma Research BIO [1]. The factory was designed in accordance with KFDA (MFDS) GMP guidelines and has a production capacity of approximately 2 million vials per year [3]. According to PRP, the addition of ReNTox to the company’s portfolio has complemented its line of aesthetic products and strengthened its position in the international market [4].
Formulation and dosage form
ReNTox contains active botulinum toxin type A, with one vial typically providing 100 IU (a 200 IU version is also available). The product is supplied as a lyophilized white powder to be dissolved in saline. Each 100-unit preparation also contains 0.5 mg of human albumin and 0.9 mg of sodium chloride as excipients. The manufacturer emphasizes that the ReNTox formula is very pure; it does not contain gelatin or additional foreign proteins (only stabilizers), which is intended to reduce the risk of immune reactions. After adding the diluent (0.9% saline), the powder dissolves into a colorless, clear solution ready for injection.
Doses and packaging
ReNTox is sold in single-use vials of 100 IU (sometimes also 200 IU) intended for administration after dilution. According to the recommendations, 100 IU is dissolved in 2.5 ml of saline, resulting in a concentration of 4 IU in 0.1 ml of solution [5]. The product is usually supplied with an insulin syringe and a saline ampoule (e.g., 20 ml), although these parameters may vary depending on the supplier. The diluted solution should preferably be used within 24 hours; unopened vials should be stored in a refrigerator (2–8°C). ReNTox has a shelf life of approximately 36 months from the date of manufacture.
Aesthetic applications
ReNTox is primarily intended for smoothing facial expression lines. It is mainly used on forehead wrinkles, frown lines (between the eyebrows), and crow’s feet around the eyes [6]. Just 3 days after the injection, most patients notice a reduction in the depth of their wrinkles, and the effect lasts for a long time (see below). In aesthetic practice, ReNTox is used in the same way as other botulinum toxins (injected into the appropriate facial muscles), which allows for a natural skin smoothing effect without surgery [6]. The product has gained the trust of clinics in Korea and Japan thanks to its effectiveness and patient satisfaction; according to the manufacturer, the effects are visible after just 3 days and last up to 6–8 months.
Medical applications
In addition to aesthetic applications, ReNTox also has a number of therapeutic indications typical for botulinum toxins. These include blepharospasm, hemifacial spasm, cervical dystonia (wry neck), and focal spasticity after stroke. In addition, the product can be used to treat paralytic strabismus or limb spasticity in children after stroke or cerebral palsy. ReNTox is indicated for the relief of involuntary muscle spasms (it helps both in neurological movement disorders and in the reduction of expression lines). Its broad spectrum of action means that it is also used in other conditions requiring muscle blockage, similar to classic botulinum preparations.
Comparison with other preparations
ReNTox belongs to the so-called “premium” botulinum toxins, distinguished by its very high purity and minimal diffusion of the drug in tissues. In its form, ReNTox is free of additional proteins (albumin is the only stabilizer), which gives it hypoallergenic properties and limits the formation of neutralizing antibodies. The manufacturer emphasizes that the effect appears quickly (often after 2–3 days) and lasts longer than with some other preparations (6–8 months). Thanks to its lower diffusion, ReNTox is considered safe for precise treatments around the eyes and forehead. In clinical trials (published by sellers), ReNTox achieves results in reducing wrinkle depth comparable to other type A toxins (e.g., effectiveness in reducing frown lines in 80–85% of patients). In medical practice, there are no significant differences in the side effect profile between ReNTox and its competitors – only temporary swelling or redness at the injection site is typical.
Action and duration of effect
ReNTox blocks the release of acetylcholine in muscle synapses, leading to temporary paralysis (relaxation) of the affected facial muscles. Complete smoothing of wrinkles usually occurs within 1–2 weeks after injection. According to the manufacturer, the therapeutic effect is sometimes noticeable as early as 3 days after administration. In most patients, the effect lasts for a long time – usually from about 4 to 8 months. After this time, the neuromuscular connections in the neurons are rebuilt, so another injection is necessary. The mechanism of action of ReNTox and the course of the effect are essentially the same as for other type A botulinum toxins, i.e., reversible and repeatable with each subsequent dose.
Clinical trials and safety
Although it is difficult to find independent publications on ReNTox in the scientific literature, the product has undergone internal quality testing and meets regulatory standards. The modern production facility in Gangneung complies with the Korean MFDS (KFDA) guidelines for good manufacturing practice (GMP)[3], which means rigorous control of the purity and safety of the preparation. Clinical experience indicates that ReNTox has a side effect profile similar to other toxins: the most common are temporary local effects (pain, swelling at the injection site) and transient muscle weakness (e.g., the eye “drops” briefly). Thanks to the use of a minimal amount of inactive proteins (no proteins other than albumin), the preparation has a reduced immunogenic potential and a lower risk of developing immunity compared to traditional botulinum formulas.
Expert opinions and legal regulations
ReNTox does not yet have a separate EMA authorization for the European market, which is why it is mainly used in the EU on the basis of parallel imports or by private aesthetic clinics. Aesthetic medicine experts point out that ReNTox produces effects similar to those of well-known brands (e.g., in Korea, it is sometimes referred to as a competitor to Botox), but is often offered at a more favorable price. It enjoys positive reviews due to its fast action and long-lasting effects – many doctors praise its high purity and stability of effect above all else.
